I think if I were buying a retrogameboyz controller as you have pictured above, then I'd go with a right hand stick.  Because that's how I play using a real 5200 controller -- right hand on stick. 

I think it was the Vectrex that was the first "left stick" that I can think of.  And then the Gameboy/NES kind of set the standard for left pad that still exists today. 

For me, I try to get used to however the original controller works for a particular system.  I have no real "general" preference.  It changes system to system.

I'm right hand dominate but all of my joystick like controllers are left handed. Not really sure why, other than that is how many arcades of the day were when I was a kid, and also because it seems to me that actual arcade controllers that were released for home use in the 80s/90s were also left handed. Like the Sega arcade controller for the Genesis for instance. So to me it just seems I prefer that when using an actual joystick. 

Although of course when I was kid I held my CX-40s with the base in my left hand and using my left thumb for the fire button with my right hand actually moving the joystick. Same with Colecovision and Intellivision controllers. I hold the base of the controller with my left using my thumb and index for the buttons and my right hand for the control.

But yeah for some odd reason when it comes to larger controllers like what is shown above, I go with the left handed model and actually don't like the way it feels using the right handed versions.

Of course modern gaming has us all moving our characters mainly with the left hand and using the right for buttons and camera control. Kinda that way on PCs with mouse/keyboard combos too for most of us. Using the left hand for the WASD keys and the right hand for the mouse and fire.
